This is a multistep project requiring the following steps: 1. Evaluate whole genome sequencing data from a cohort of 2500 previously sequenced individuals to isolate those that carry heterozygous mutations in genes that are functionally annotated with the GO term "XYZ." 2. Assess these genes, using RNA-Seq data in the publically available Allen BrainAtlas: goal in this step is to isolate those genes that are involved in neuronal function as such. This will require a differential expression analysis either across different locations of the brain or compare with RNAseq data from other tissues. Genes that do not show brain expression, and genes that do not show evidence for involvement in neuronal function, are excluded. 3. For the cohort of genes that proceed to this step, utilize mutation programs such as SIFT or POLYPHEN and the DGV (Database of Genomic Variants) to establish pathogenicity of the mutations. 4.
